General description
LIM domain-containing protein 2 (UniProt Q9BT23) is encoded by the LIMD2 (also known as SB143) gene (Gene ID 80774) in human. The LIM domain is organized as a tandem zinc-finger structure that functions as a modular protein-binding interface and LIM domain-containing proteins play diverse roles in gene expression, cyto-architecture, cell adhesion, cell motility, and signal transduction. Human LIM domain-only (LMO) proteins are involved in many normal developmental processes, as well as in the onset and progression of several cancers, including T-cell leukemia, breast cancer, and neuroblastoma. LIMD2 is a LMO protein that is highly expressed in papillary thyroid carcinoma (PTC) lymph node metastasis (LNM), but absent from the primary tumor or normal thyroid tissue in matched PTC patient samples. LIMD2 regulates cell motility and affects tumor progression via its role in the integrin-linked kinase (ILK) pathway. LIMD2 is reported to activate ILK activity via direct binding to ILK kinase domain near the active site, LIMD2-knockdown in cancer cells results in reduced cell migration. Likewise, epitopic LIMD2 expression promotes the migration of ILK-expressing fibroblasts, but not fibroblasts lacking ILK.
