General description
Mitogen-activated protein kinase kinase kinase 5 (MAP3K5), also known as, MAPK/ERK kinase 5 (MEK Kinase 5; MEKK 5) or Apoptosis signal-regulating kinase 1 (ASK-1) protein, is a serine/threonine kinase that is part of the MAP kinase signal transduction pathway. The MAP kinase pathway is central to several signaling cascades the help determine everything from cell fate in differentiation to cell survival in apoptosis. MAP3K5 is critical for innate immune response against numerous pathogens and MAP3K5 also mediates cells responses to various cellular stresses such as oxidative stress, and responses mediated by inflammatory signaling agents such as TNF or LPS. Moreover, MAP3K5 has has been found to be involved in cancer, diabetes, cardiovascular and neurodegenerative diseases. MAP3K5 is membrane associated though not membrane bound directly. Its expression thus is primarily endoplasmic reticulum but can become strongly perinuclear under some conditions. Found in most tissues, it is abundant in heart and pancreas. It′s activity is regulated via phosphorylation-dephosphorylation processes as well as though its interactions with a many other protein partners to form numerous multimolecular cell signaling complexes. EMD-Millipore’s Anti-MAP3K5 mouse monoclonal, clone 2E4, has been tested in western blotting against recombinant human MAP3K5 protein, and in paraffin-embedded immunohistochemistry with human cancer tissues, and in immunofluorescent immunocytochemistry, and flow cytometry on human HeLa cells in culture.
