General description
Interferon-induced protein with tetratricopeptide repeats 5 (UniProt Q13325; also known as IFIT-5, Interferon-induced 58 kDa protein, P58, Retinoic acid- and interferon-inducible 58 kDa protein) is encoded by the IFIT5 (also known as ISG58, RI58) gene (Gene ID 24138) in human. IFIT1, IFIT2, IFIT3, and IFIT5 constitute the family of interferon-stimulated genes (ISGs) that are induced in response to virus infection, stimulation by interferons (IFNs) or double-stranded RNA (dsRNA). The protiens encoded by IFIT1 and IFIT2 are called ISG56 (p56) and ISG54 (p54) in both human and murine species, while the murine IFIT3 (ISG49 or p49) is of a different size than its human counterpart (ISG60 or p60) and no known murine IFIT5 analog exists. IFIT family proteins restrict virus replication by various means. IFIT1 is known to disrupt complex formation between eIF3 and HCV RNA and inhibit HPV E1 helicase. IFIT3 is shown to enhance TBK1 and MAVS interation to potentiate antiviral response. IFIT5 is reported to facilitate the interaction between I B kinase (IKK) and TGF- -activated kinase 1 (TAK1) by binding both proteins, resulting in enhanced NF- B activation as a reuslt of increased IKK phosphorylation and activation. In addition, both IFIT1 and IFIT5 are shown to recognize viral RNA carrying 5 -triphosphate (PPP-RNA). Human IFIT5 contains eight tetratricopeptide repeat (TPR) helix-turn-helix motifs (a.a. 51-84, 94-127, 138-173, 181-214, 249-282, 338-371, 376-410, 435-468), with a.a. 254-260 within TPR repeat 5 mediating PPP-RNA interaction.
