General description

GST-tagged recombinant full-length human SAMHD1.
~72 kDa observed. 72.2 kDa (isoform 1), 69.4 kDa (isoform 2), 63.7 kDa (isoform 3), 68.1 kDa (isoform 4) calculated.

Application

Flow Cytometry Analysis: A representative lot detected a downregulated SAMHD1 immunoreactivity among cutaneous T cell lymphoma (CTCL) patients-derived CD4+ PBMCs when comparing to CD4+ PBMCs from healthy donors (de Silva, S., et al. (2014). J. Invest. Dermatol. 134(2):562-565).
Flow Cytometry Analysis: A representative lot detected SAMHD1 immunoreactivity in CD123+ plasmacytoid dendritic cells (pDCs) from human PBMCs and a downregulated SAMHD1 immunoreactivity in pDCs when cocultivated with PHA–IL-2-activated CD4+ T cells (Su, B., et al. (2014). J. Virol.88(18):10975-10981).
Flow Cytometry Analysis: A representative lot detected SAMHD1 immunoreactivity in non-infected monocyte-derived dendritic cells (MDDCs) and a greatly reduced SAMHD1 immunoreactivity in MDDCs exposed to virion-like particles (VLPs) generated from wild-type SIV-3 (simian immunodeficiency virus type 3)-positive plasmid (Puigdomènech, I., et al. (2013). J. Virol.87(5):2846-2856).
Western Blotting Analysis: A representative lot detected SAMHD1 in lysates from non-infected monocyte-derived dendritic cells (MDDCs), but not in MDDCs exposed to virion-like particles (VLPs) generated from wild-type SIV-3 (simian immunodeficiency virus type 3)-positive plasmid (Puigdomènech, I., et al. (2013). J. Virol.87(5):2846-2856).
Immunocytochemistry Analysis: 2.0 µg/mL from a representative lot detected SAMHD1 in THP-1 cells.
Western Blotting Analysis: A representative lot detected a downregulation of intracellular SAMHD1 in primary human dendritic cells (DCs) following infection by virion-like particles (VLPs) generated from wild-type SIV-3 (simian immunodeficiency virus type 3)-positive plasmid (Courtesy of Dr. Olivier Schwartz, Institut Pasteur, Paris, France).
Immunocytochemistry Analysis: 1.0 µg/mL from a representative lot detected SAMHD1 expression in THP1 human monocytic cells by fluorescent immunocytochemistry (Courtesy of Dr. Olivier Schwartz, Institut Pasteur, Paris, France).
Flow Cytometry Analysis: 0.4 µg/mL from a representative lot detected a downregulation of intracellular SAMHD1 in primary human dendritic cells (DCs) following infection by virion-like particles (VLPs) generated from wild-type SIV-3 (simian immunodeficiency virus type 3)-positive plasmid (Courtesy of Dr. Olivier Schwartz, Institut Pasteur, Paris, France).
Flow Cytometry Analysis: A representative lot detected a downregulated SAMHD1 immunoreactivity among CD4+ PBMCs 5 days post infection by a laboratory-adapted HIV-2 strain (ROD) or by two other HIV-2 isolates (TOE and ROK) from infected patients (Chauveau, L., et al. (2015). Retovirology. 12:2).

Physical form

Purified mouse monoclonal IgG1κ antibody in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.

Preparation Note

Stable for 1 year at 2-8°C from date of receipt.

Analysis Note

Evaluated by Western Blotting in THP-1 cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected SAMHD1 in 10 µg of THP-1 cell lysate.
