General description
Antigen peptide transporter 2 (UniProt Q03519; also known as APT2, ATP-binding cassette sub-family B member 3, Peptide supply factor 2, Peptide transporter involved in antigen processing 2, Peptide transporter PSF2, Peptide transporter TAP2, PSF-2, Really interesting new gene 11 protein) is encoded by the TAP2 (also known as ABCB3, PSF2, RING11, Y1) gene (Gene ID 6891) in human. TAP2 (ABCB3) belongs to the subfamily B of ATP-binding cassette (ABC) transporters, members of which also include TAP1 (ABCB2) and multidrug resistance protein 1 (MDR1, ABCB1, P-gp). TAP is a heterodimeric peptide transporter composed of TAP1 and TAP2 that dimerize via their nucleotide-binding domain (NBD; a.a. 503-510) in a head-to-tail fashion. TAP preferentially mediates the transport of 8- to 16-a.a. peptides into the ER in a process powered by ATP-binding and hydrolysis. TAP1 and TAP2 harbor a specialized N-terminal transmembrane domain (TMD0) that is followed by the TAP half-transporter core module composed of one TMD and one NBD. TMD0 is composed of a 4-TM bundle that serves as the interaction platform for the assembly of the peptide-loading complex (PLC).
