General description
T-complex protein 1 subunit beta (TCP-1-beta) or alternatively known as CCT-beta which is encoded by the human gene named CCT2 or 99D8.1, is a protein that plays a role as a molecular chaperone. This means that CCT2 assists in the non-covalent folding or unfolding and the assembly or disassembly of other macromolecular structures, but does not occur in these structures when the structures are performing their normal biological functions. In the case of CCT2 it assists in the folding of various proteins including actin and tubulin, two very important cytoskeletal proteins in all cells. CCT2 is cytoplasmically expressed in mammalian cells, and CCT2 in worms at least, CCT2 is expressed in most, if not all, tissues. Moreover via inhibition assays in worms, CCT2 is required for a variety of functions including normal transgene repression and subcellular localization events, embryonic osmotic integrity, embryonic and larval viability, fertility, vulval development, and body color. In humans, CCT2 is also a component of a huge multi-protein complex called the BBS/CCT (BBSome) complex which is composed of several proteins named MKKS, BBS10, BBS12, TCP1, CCT3, CCT4, CCT5, and CCT8, that forms part of the cilia. It is thought that CCT2 may play a role in the assembly of the BBSome which is needed for ciliogenesis, and CCT2 appears to regulate the transport of vesicles to the cilia. EMD-Millipore’s Anti-CCT2 mouse monoclonal antibody has been tested in western blot against recombinant protein as well as cell lysates from HeLa, MCF-7, Jurkat, T47, K562, A431, NIH3T3, PC-12 and COS7 cells. The antibody has also been successfully tested in fluorescent immunocytochemistry on 3T3-L1 cells and in flow cytometry on NIH3T3 cells in culture and in direct ELISA on varying concentrations of recombinant antigen.
