BODIPY-FL-C12 is a fluorescent fatty acid analog, resemble the length of C16 FA such as palmitic acid, designed for real-time visualization of lipid metabolism in living cells. Structurally, a C12 alkyl chain directs the molecule into cellular fatty acid transport pathways, while the core BODIPY fluorophore provides exceptionally bright and photostable green fluorescence (λex/λem ≈ 505/515 nm). Following rapid cellular uptake, BODIPY-FL-C12 is esterified by acyl-CoA synthetase in the endoplasmic reticulum and subsequently trafficked for robust, irreversible accumulation within the neutral lipid core of lipid droplets. This metabolic trapping mechanism generates a high signal-to-noise ratio, enabling high-resolution tracking of fatty acid uptake, transport, and storage dynamics. Its proven utility in live-cell imaging, flow cytometry, and in vivo models like C. elegans supplants traditional endpoint assays.
