General description

ESGROマウスLIF培地サプリメントは、ロット間で一貫した性能が得られるように、活性ユニット/mLで供給されます。 ESGROサプリメントの各ロットについては、マウス多能性幹細胞の分化を高い信頼性で確実に阻害できるように、厳格な品質管理試験を実施しています。 ESGROマウスLIFサプリメントは、マウス胚性幹細胞（ES）および人工多能性幹細胞（iPS）のフィーダーベースおよびフィーダーフリー培養に使用できます。
マウス白血病抑制因子（LIF、mLIF）は、自然発生的な分化を抑制することにより、多能性マウス胚性幹細胞（ES）および人工多能性幹（iPS）細胞の自己複製および長期維持を促進します。

Application

ESGROサプリメントの用途としては、マウスES細胞の多能性表現型をin vitroで維持するための試薬としての使用などがあります。

推奨されるプロトコル：

ES細胞：D3およびMBL-1多能性ES細胞培養では、幹細胞表現型を有するES細胞を維持するために、組織培地1.0 mL当たり1000 UのESGRO サプリメントが必要であることが明らかになっています。 遺伝子改変ES細胞（E3）の生殖細胞系伝達にも同様の濃度のmLIFが用いられています。

推奨濃度では、10.0 Lの組織培地に対して107ユニットのESGRO サプリメントが十分量であり、 1.0 Lの組織培地に対して106ユニットのESGRO サプリメントが十分量です。
マウスES細胞培養用のESGRO白血病阻止因子（LIF）添加剤 各バイアルには10^6 units/mLが含まれています。

Physical form

ESGROサプリメントは、安定性のための担体として1% w/vウシ血清アルブミン（BSA）を含むリン酸緩衝生理食塩水1.0 mL中の106 Unitsとして液体で供給されます。
フォーマット：精製

Preparation Note

ESGRO®は、0.22ミクロン無菌ろ過されて供給され、マイコプラズマ試験陰性であることが確認されています。
ESGROサプリメントは常温で出荷されます。顧客リレーションのために保冷剤を入れることもありますが、実際には不要です。開発段階では、ESGROサプリメントを用いた広範な安定性試験を実施しています。 この製品は、濃縮状態および滅菌組織培地で希釈した状態で製造日から少なくとも18か月間は安定であり、ES細胞に対する活性が損なわれることはありません。 長期保存には、ESGROサプリメントを4°Cで保存することをお勧めします。 凍結融解は力価を低下させます。使用前にESGROサプリメントを滅菌組織培地で希釈し、便利な濃度に分注してから4°Cで保存することをお勧めします。凍結-解凍は避けてください。ES細胞培養中、製品は37°C，5%CO2インキュベーターで7日間以上安定です。

Analysis Note

比活性：ESGRO®サプリメントはマウス胚性幹細胞（D3、MBL-1）およびマウスM1骨髄性白血病細胞の両方で評価しています（4）。標準である50ユニットは、M1コロニーの50%の分化を誘導する組織培地1.0mL中のESGROサプリメントの濃度として定義されています（4）。 106ユニットは、約10 μgの純タンパク質に相当し、1LのES細胞培地を十分に処理できます。

胚性幹細胞アッセイ：1000 units/mLのマウス骨髄性白血病での分化阻害、M1アッセイ：比活性 >/= 108 units/mg
