General description

5,5′-ジチオビス（2-ニトロ安息香酸）は、DTNBまたはエルマン試薬とも呼ばれます。水溶性の試薬であり、チオール基と反応します。中性pHで用いることができるため便利です。チオラートアニオン（RS-）はエルマン試薬（DTNB2-）と反応し、2-ニトロ-5-チオ安息香酸アニオン（TNB2-）および混合ジスルフィド（R-S-TNB-）を与えます。TNB2-は高いモル吸収係数を持ち、412 nmの波長を吸収します。この吸収はpH 7.3以上ではpHの影響を受けません。

Application

エルマン試薬はアセチルコリンエステラーゼ測定、およびシステイン残基の特異的標識のために用いられることがあります。タンパク質、ペプチドおよび組織中の遊離スルフヒドリル基含量測定のための高感度試薬です。
