General description

ラミニンは、分子量がかなり大きい、非コラーゲン性の接着性糖タンパク質です。ヘテロ三量体タンパク質であり、α、β、γ鎖として知られるジスルフィド結合した3つのポリペプチドを有しています。ラミニンの複数のポリペプチドドメインは、複数のインテグリン二量体、ジストログリカンとシンデカン、その他のECM成分などの、複数の細胞表面受容体に結合できます。この細胞外マトリックスマルチドメイン三量体糖タンパク質が、接着、増殖、および分化をサポートする基底膜の主成分です。

Application

マウスのエンジェルブレス−ホーム−スワーン腫瘍基底膜由来のラミニンは以下に使用されています：
  • インスリン産生細胞（IPC）の分化。
  • 感染阻害アッセイ。
  • 増殖アッセイ。
ラミニンは、上皮、内皮、神経、筋、および肝細胞を含む、多くの細胞タイプの増殖および分化をサポートします。 細胞培養足場として、1～2 μg/cm2で使用することをお勧めします。 最適な濃度は、アプリケーションや研究目的だけでなく、細胞の種類によっても異なります。

Biochem/physiol Actions

ラミニンタンパク質は、動物組織における構造的足場に不可欠の成分です。 ラミニンは、エンタクチンやパールカンを介してタイプIVコラーゲンと会合し、インテグリン受容体、ジストログリカン糖タンパク質複合体、およびLutheran血液型糖タンパク質を通じて細胞膜に結合します。 ラミニンは、コラーゲンとの結合、細胞接着、ヘパリンとの結合、および神経突起伸長のための活性ドメインを持っています。

Preparation Note

本製品は、50 mM Tris-HCl、150 mM NaClの1～2 mg/mL溶液として提供されます。 0.2 μmフィルターでろ過されています。ゲルの形成を避けるために、使用前に2～8℃でゆっくりと溶液を溶解してください。 コーティングに使用する場合は、緩衝塩溶液で希釈して、最小量で培養表面をコートし、37℃で2時間インキュベートしてください。 細胞をプレーティングする前に、PBSで3回洗浄してください。ラミニンコーティングは、2～8℃で1か月保存できます。
