General description
核内タンパク質抽出法の手順により、低浸透圧緩衝液を用いて細胞を膨潤させることができます。細胞は次に破壊され、細胞質分画が除去され、そして核内タンパク質が、高塩濃度の緩衝液により核から放出されます。

本キットでは、哺乳動物の組織または培養細胞から核抽出物または細胞質タンパク質抽出物を得るための完全なシステムを提供しています。抽出に必要な試薬がすべて含まれています。
詳細なtechnical bulletinにさまざまな手順が記載されており、特定のアプリケーションに最も適したものを選択することができます。 例として、核タンパク質には界面活性剤を用いた抽出法と界面活性剤を用いない抽出法から選択し、大部分の細胞タイプには低張溶解バッファー、壊れやすい細胞には等張溶解バッファーを選択してください。 さらに本キットにより、希釈バッファーを用いて核抽出液の塩濃度を低下させる手順を行うことができます。 NuCLEARを必要に応じて柔軟に使用することで、最適なタンパク質抽出が可能です。 2時間未満で抽出液を調製することができ、核抽出液と細胞質抽出液の間で交差汚染することがほとんどないため、抽出液の純度は非常に高いです。
