General description

西洋ワサビペルオキシダ-ゼは西洋ワサビ(Amoracia rusticana)の根から単離された酵素で、ペルオキシダ-ゼのフェロプロトポルフィリン類に属します。HRPは4個のジスルフィド架橋結合を有する1本のポリペプチドです。18%の糖を含む糖タンパク質です。糖の組成はガラクト-ス、アラビノ-ス、キシロ-ス、フコ-ス、マンノ-ス、マンノサミン、ガラクトサミンからなり、個々のアイソザイムによって異なります。 この酵素の分子量(約44 kDa)には、ポリペプチド鎖(33,890 Da)、ヘミン+Ca2+(約700 Da)、糖(約9,400 Da)が含まれます。HRPには少なくとも7種類のアイソザイムが存在します。 西洋ワサビペルオキシダ-ゼアイソザイムの等電点は3.0~9.0です。

Application

ホースラディッシュペルオキシダーゼ（HRP）は、セイヨウワサビの根から単離されました（Amoracia rusticana）。 ウェスタンブロット、ELISA および免疫組織化学などの生化学アプリケーションに使用されています。 ホースラディッシュペルオキシダーゼは、タンパク質などの標的分子の弱いシグナルを増幅させその検出可能性を向上させます。 ホースラディッシュペルオキシダーゼ製品である P8250 は、炎症歯肉における非経口抗原の研究およびエボラウイルスの糖タンパク質毒性の研究に使用されています。
本酵素は、成熟した Tall Fescue の葉身抽出物のペルオキシダーゼアッセイにおいて比較に使用されています。また、H2O2 産生の測定にも使用されています。

Biochem/physiol Actions

ホースラディッシュペルオキシダーゼを基質とインキュベーションすると、標的分子の着色、蛍光または発光誘導体が生成し定量が可能となります。 ホースラディッシュペルオキシダーゼは、cydAB 変異体において阻害レベルをわずかに低減させることが示されています。
HRP は過酸化水素（H2O2）と容易に結合し、その生成物 [HRP-H2O2] 複合体は幅広い種類の水素供与体を酸化することができます。至適 pH は 6.0～6.5 で、酵素は 5.0～9.0 の pH 範囲で最も安定です。HRP は、グルタルアルデヒド、過ヨウ素酸塩酸化、ジスルフィド結合を含むいくつかの異なる方法で抗体コンジュゲートを形成することが可能であり、アミノおよびチオール架橋剤も使用できます。β-ガラクトシダーゼおよびアルカリホスファターゼ酵素標識よりも小さく安定です。このため最も望ましい標識となっています。また、グリコシル化により非特異的結合がさらに低下します。溶液中のグルコースおよび過酸化物の同定にも使用されます。アジ化ナトリウム、シアン化物、L-シスチン、二クロム酸塩、エチレンチオウレア、ヒドロキシルアミン、硫化物、バナジン酸、p-アミノ安息香酸および　Cd2+、Co2+、Cu2+、Fe3+、Mn2+、Ni2+、Pb2+ イオンは酵素活性を阻害することが見出されています。

Analysis Note

RZ(Reinheitszahl)値は、0.5~1.0 mg/mL脱イオン水溶液で測定したA403/A275の吸光度比です。これはヘミン含量を測定するもので、酵素活性を測定するものではありません。RZ値の高い標品でも酵素活性が低いことがあります。
予備試験では、少なくとも5種類のアイソザイムが含まれることが示されています。

Other Notes

1ピロガロールユニットは、pH 6.0、20°Cで 20秒あたり1.0 mgのプルプロガリンをピロガロールから生成する酵素量です。
