      Revision22-September-2011 RFH
      SynonymsProcollagen C-proteinase, PCP
      Application Data
*Adjusted for Substrate Blank **Derived using calibration standard MCA-Pro-Leu-OH
DescriptionRecombinant, human bone morphogenetic protein (BMP-1) consisting of amino acids 121-730 and expressed in NS0-derived mouse myeloma cells with a C-terminal His•Tag® sequence.
      FormSolid
FormulationLyophilized from 0.2 µm filtered HEPES, NaCl, CaCl₂, ZnCl₂, and Brij®35 detergent.
      Recommended reaction conditions

      Activity Assay Protocol:
      Materials:
      Assay Buffer: 25 mM HEPES, 0.1% Brij®35 detergent (v/v), pH 7.5
      Recombinant human BMP-1
      Fluorogenic Peptide Substrate: MCA-Tyr-Val-Ala-Asp-Ala-Pro-Lys( DNP)-OH
      F16 Black Maxisorp Plate
      Fluorescent Plate Reader

      Assay:
      1. Dilute rhBMP-1 to 20 ng/µl in Assay Buffer.
      2. Dilute substrate to 20 µM in Assay Buffer.
      3. Add 50 µl of diluted rhBMP-1 to designated wells of the black plate. Start the reaction by adding 50 µl diluted Substrate. Include a Substrate Blank that contains only Assay Buffer (in place of rhBMP-1).
      4. Read at excitation and emission wavelengths of 320 nm and 405 nm (top read), respectively, in kinetic mode for 5 min.
      5. Calculate specific activity:
