General description

Amyloid Precursor Protein (APP) is expressed in the brain, kidney, heart, and spleen of fetal tissues; it is induced during neuronal differentiation. The most-substantiated role for APP is in synaptic formation and repair; its expression is upregulated during neuronal differentiation and after neural injury. In adult brain, highest expression of APP is found in the frontal lobe of the cortex. Moderate expression is observed in the cerebellar cortex, the posterior perisylvian cortex-opercular gyri and the temporal associated cortex. Defects in APP are a cause of autosomal dominant Alzheimer′s disease (AD). Deposits of amyloid protein in senile plaques near nerve processes are found in the brains of aged humans and in cases of Alzheimer′s disease. The principle component of this extracellular amyloid is beta/A4, a 4 kDa peptide derived from the larger APP. The creation, transport and function of these proteins are currently under investigation.

Analysis Note

Evaluated by immunohistochemistry(paraffin) on Alzheimer’s Disease-Hypothalamus Tissue.

Immunohistochemistry(paraffin): Amyloid (β-A4-protein, cat. # MAB348) staining on Alzheimer’s Disease-Hypothalamus. Tissue pretreated with Citrate, pH 6.0. The antibody was diluted to 1:80, using IHC-Select® Detection with HRP-DAB. Immunoreactivity is seen as staining on plaque deposits (dark brown) or as cerebral vascular amyloid deposition in the walls of blood vessels (brown).

Other Notes

Immunohistochemistry: Previous lot tested Immunohistochemistry on 4% paraformaldehyde/15% picric acid perfused and fixed, frozen and paraffin embedded sections. Optimal working dilutions must be determined by the end user.
