DescriptionRabbit polyclonal antibody supplied as undiluted serum. Recognizes the ~60 kDa PDK1 protein.
Background3-phosphoinositide-dependent kinase 1 (PDK1; accession no. NP-002613) is a serin/threonine protein kinase that plays a major role in cell signaling pathways, cancer, cell growth, and tumorigenesis. PDK1 consist of a N-terminal kinase domain and a C-terminal pleckstrin homology (PH) domain. The PH domain binds to the PI 3-K products, PIP3 and PIP2, which target PDK1 to the plasma membrane where it phosphorylates AKT at Thr308. PDK1 can also directly associate and phosphorylate many other important substrates including PKC, p70S6K, and SGK. PDK1 is activated by phosphorylation at Ser241. Recently, an autonomous nuclear-specific PI 3-K signaling pathway has been discovered that contains all of the key PI 3-K signaling components including AKT, PKC, and p70S6K. PDK1 has also been found in the nucleus, suggesting that this protein is shuttled from the cytoplasm to the nucleus.
