      OverviewThe SMC® Human PD-1 High Sensitivity Immunoassay Kit contains all reagents required to perform the assay. This assay takes advantage of a standard immunoassay workflow configured in a standard plate format. Assay protocols are similar to existing sandwich ELISA methods with two key differences: 1) Elution buffer disrupts the sandwich, separating the labeled detection antibody for quantification. 2) The SMCxPRO® immunoassay systems detect analytes using Single Molecule Counting (SMC®) technology.
      Background Information
      The SMC® Human PD-1 High Sensitivity Immunoassay Kit utilizes Single Molecule Counting technology, based on the trusted sandwich immunoassay principle, to digitally measure Programmed Cell-Death Protein 1 (PD-1) in Human Serum and Plasma samples. A capture antibody specific for Human PD-1 has been pre-coated onto paramagnetic microparticles (beads). The user pipettes beads, standards, and samples into uncoated microplate wells. During incubation, the PD-1 present in the sample binds to the capture antibody on the coated beads. Unbound molecules are washed away during the subsequent wash steps. Fluor-labeled detection antibody is added to each well and incubated. This detection antibody recognizes and binds to PD-1 that has been captured onto the beads, thus completing the immunosandwich. Elution buffer is then added and incubated. The elution buffer dissociates the bound protein sandwich from the beads surface releasing the labeled antibodies. These antibodies are separated during transfer to a 384-well read plate. The plate is loaded into the SMCxPRO® System where the labeled molecules are detected and counted. The number of fluor-labeled detection antibodies counted is directly proportional to the amount of PD-1 present in the sample when captured. The amount of PD-1 in unknown samples is interpolated from a standard curve.

      Application NotesProgrammed cell death protein 1 (PD-1), CD279, is a type I membrane protein found in T cells, B cells, and macrophages that binds Programmed cell death ligand 1 and 2 (PD-L1 and PD-L2). PD-1 is found in both soluble and insoluble forms. PD-1 regulates the body’s immune response through a variety of pathways, down-regulating immune system activity and suppressing T-cell inflammatory activity. PD-1 has been implicated as playing a role in cancers, autoimmune diseases, the immune response to viruses, and in immunological pathways related to brain repair.
