This cationic lipid demonstrates superior performance in vivo expression and tolerability, particularly for hepatic and pulmonary delivery applications. Formulations with CP-LC-0729 have a measured pKa of ~6.78 and promotes effective endosomal escape, an essential mechanism for successful mRNA, saRNA, or circRNA release into the cytosol. Its branched hydrophobic structure supports the formation of stable, homogenous LNPs with high encapsulation efficiency and low polydispersity.
