MN714 is a cell-permeable pivaloyloxymethyl (POM) ester precursor of MN551, an E3 ligase suppressor of cytokine signaling 2 (SOCS2) inhibitor. Upon entering cells, MN714 is hydrolyzed by cellular esterases to MN551 that covalently modifies SOCS2 SH2 domain EF loop Cys111. MN551 also effectively modifies SH2 containing protein (CISH) (Cys114), but not SOCS4 (Cys350) or SOCS6 (no EF loop Cys). MN714 treatment of K562 cells (10 µM for 6h) prior to lysate preparation compeletly abrogates endogenous SOCS2 binding of immobilized GHR pY595 substrate peptide.
