General description

This reagent is a modified lipid containing an omega-terminal alkyne. The terminal alkyne group can be used in a highly specific linking reaction with azide-containing reagents, known as ‘click chemistry′, in the presence of a copper (Cu)-containing catalyst.
Alkyne-cholesterol is a clickable cholesterol probe proprietary for Avanti Polar Lipids. They differ from cholesterol by having an alkyne group.

Biochem/physiol Actions

Alkyne-cholesterol is not applicable for use in living cells due to copper ion (Cu (I) and Cu (II)) requirements. It aids in easy and sensitive detection of cholesterol using various biophysical methods. Alkyne-cholesterol can be conjugated to boron-dipyrromethene (BODIPY) fluorescent dye. It is useful in cholesterol trafficking studies in various organelles including mitochondria, endoplasmic reticulum and Golgi.

Cholesterol transport between cellular organelles comprised vesicular trafficking and nonvesicular exchange; these processes are often studied by quantitative fluorescence microscopy. A major challenge for using this approach is producing analogs of cholesterol with suitable brightness and structural and chemical properties
