P4HA1 (prolyl 4-hydroxylase, α polypeptide I) gene encodes an α subunit of prolyl 4-hydroxylase, an enzyme that catalyzes post-translational formation of 4-hydroxyproline from proline residues in -Xaa-Pro-Gly- sequences during collagen synthesis. It facilitates the proper three-dimensional folding of procollagen chains that are synthesized newly to form stable triple helical molecules. The enzyme is composed of two identical α subunits and two β subunits. P4HA1 and P4HA2 are essential for collagen deposition. These are involved in the remodeling of ECM composition, alignment, and mechanical properties in response to hypoxia along with another enzyme PLOD2 (lysyl hydroxylase). The expression of P4HA1 is induced by hypoxia-inducible factor 1 (HIF-1).
