Application CommentsDoes not cross-react with full length caspase-9 or other cleaved caspases. For immunoblotting, incubate membrane with antibody diluted in 5% nonfat dry milk, 1X TBS, and 0.1% Tween®-20 detergent at 4°C overnight with gentle shaking. Antibody should be titrated for optimal results in individual systems.

DescriptionProtein A and immunoaffinity purified rabbit polyclonal antibody. Recognizes the ~35 kDa large subunit of caspase-9.
BackgroundCaspase-9 is an important member of the cysteine aspartic acid protease (caspase) family. Upon apoptosis cytochrome c released from mitochondria associates with procaspase-9 (APAF, ~47 kDa) and this complex processes the procaspase-9 into a large active subunit (~17 or ~38 kDa) and a small subunit (~10 kDa) by self-cleavage at Asp315. The cleaved caspase-9 also processes other caspases, including caspases 3 and 7, to start a caspase cascade that leads to programmed cell death. Procaspase-9 can also be cleaved by caspase-3 at Asp330, serving as a positive sign to amplify the apoptotic signal in the caspase activation pathway.
