Gene Information

General description

Tumor necrosis factor receptor superfamily member 13B (UniProt: O14836; also known as Transmembrane activator and CAML interactor) is encoded by the TNFRSF13B (also known as TACI) gene (Gene ID: 23495) in human. Tumor necrosis factor receptor superfamily member 13B (CD267/TACI) is a type I transmembrane glycoprotein primarily expressed on B cells. CD267/TACI contains extracellular cysteine-rich domains that mediate binding to its ligands, BAFF (B-cell activating factor) and APRIL (a proliferation-inducing ligand). These interactions are crucial for the activation of downstream signaling pathways, including the canonical and non-canonical nuclear factor- B (NF- B) pathways, which regulate B-cell survival, proliferation, and immunoglobulin production. CD267/TACI is localized to the plasma membrane and can be internalized upon ligand binding. It interacts with TNF receptor-associated factor (TRAF) proteins, which serve as key mediators of its signaling. Dysregulated CD267/TACI signaling has been implicated in common variable immunodeficiency (CVID), autoimmunity, and B-cell malignancies. Mutations in TNFRSF13B are associated with defective class-switch recombination and impaired antibody responses, leading to susceptibility to infections and autoimmune conditions. Its involvement in immune regulation underscores its therapeutic potential in treating immunodeficiencies and autoimmune disorders.

Immunogen

Irradiated CD267/TACI-transfected rat basophilic leukemia (RBL) cells.

Application

Quality Control Testing

Evaluated by Flow Cytometry in human peripheral blood mononuclear cells (PBMC).

Flow Cytometry Analysis: 1.0 µg from a representative lot detected CD267/TACI in one million human peripheral blood mononuclear cells (PBMC).

Tested Applications

Flow Cytometry Analysis: A representative lot detected CD267/TACI in Flow Cytometry application (Ng, L.G., et al. (2004). J Immunol. 173(2):807-17).

ELISA: A representative lot detected CD267/TACI in ELISA (Ng, L.G., et al. (2004). J Immunol. 173(2):807-17).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Biochem/physiol Actions

Clone 1A1 is a rat recombinant monoclonal antibody that detects CD267/TACI. It targets an epitope from the extracellular domain.
