General description
C-type lectin domain family 6 member A (UniProt: Q6EIG7; also known as C-type lectin superfamily member 10, Dendritic cell-associated C-type lectin 2, DC-associated C-type lectin 2, Dectin-2) is encoded by the Clec6a (also known as CLECSF10, DECTIN2) gene (Gene ID: ) in human. Dectin-2 plays a critical role in the innate and adaptive immune system by recognizing pathogen-associated molecular patterns (PAMPs) and initiating immune responses. This receptor is an ITAM-coupled C-type lectin receptor (CLR); a single-pass type II transmembrane protein. It features a carbohydrate recognition domain (CRD) that allows it to bind specifically to high-mannose polysaccharides found on the surface of various pathogens, including fungi and some bacteria. It is characterized by a single extracellular (aa 42-209) C-type lectin-like domain (CTLD) connected by a transmembrane region (aa 22-41) to a short cytoplasmic tail (aa 1-20) that lacks a clear intracellular signaling motif. Association of Dectin-2 with adaptor proteins, such as the Fc receptor (FcR ) chain or DNAX-activation protein 12 (DAP12), is required for the engagement to Syk pathway, leading to the activation of NF- B and the production of pro-inflammatory cytokines. Its expression is particularly high in the spleen and lymph nodes and primarily expressed on dendritic cells and macrophages. Dectin-2 has been implicated in various diseases, particularly those involving fungal infections due to its role in recognizing fungal cell wall components. dysregulation of Dectin-2 signaling has been associated with inflammatory and autoimmune diseases, highlighting its importance in maintaining immune homeostasis.
