General description
Big endothelin-1 is a 39 amino acid peptide, which is synthesized and secreted by vascular smooth muscle and endothelium cells. It is produced as an inactive enzyme, and is cleaved and activated by serine and aspartic proteases and metalloproteinases. Activated form is called ET-1, which is a potent vasoconstrictor. ET-1 is also produced by the action of endothelin converting enzyme (ECE) on big-ET-1. In guinea pigs, big-ET-1 causes the release of prostanoids and thromboxanes.
Application
Big Endothelin 39 rat has been used for charting of a standard curve in the study of the role of ET-1 in ventricular dilatation.
Biochem/physiol Actions
Precursor of Endothelin-1; vasoconstrictor activity appears to be due to its conversion to endothelin 1 by endothelin converting enzyme in the tissue.

Immunocytochemical localization of big endothelin-1 (big ET-1), ET-1, and ET receptor A and B (ET(A) and ET(B)), and gene expression of prepro ET-1 mRNA were examined on the rat liver vasculature. Immunoreactivities for big ET-1 and ET-1 were preferentially seen

In porcine aortic endothelial cells, the 21-amino acid peptide endothelin-1 (ET-1) is formed from a 39-amino acid intermediate called "big endothelin-1" (big ET-1) by a putative ET-converting enzyme (ECE) that cleaves the 39-mer at the bond between Trp-21 and Val-22.

Matrix metalloproteinase-2 (MMP-2, gelatinase A) and its tissue inhibitor (TIMP-2) are mainly known for their roles in the (patho)physiological remodeling of the vasculature, angiogenesis, tissue repair, tumor invasion, inflammation, and atherosclerotic plaque rupture. A mechanism of action of MMP-2 is

All three endothelin precursor peptides, i.e. big endothelin-1 (big ET-1), big endothelin-2 (big ET-2) and big endothelin-3 (big ET-3), produced contractile responses in human isolated bronchi, demonstrating the presence of functional endothelin-converting enzyme (ECE) in this tissue. The maximal contractile
