Application

La toxina diftérica de Corynebacterium diphtheriae se ha utilizado:


  • para la merma de la microglía con objeto de estudiar el trastorno por estrés postraumático en ratones
  • inyectada intraperitonealmente en ratones transgénicos para seleccionar los híbridos después de la fusión de células madre y progenitoras hematopoyéticas trasplantadas (HSPC) durante la ablación
  • para estudiar su efecto sobre los progenitores comprometidos con el linaje de eosinófilos en una cepa de ratones con deficiencia de eosinófilos

Biochem/physiol Actions

La toxina diftérica es una toxina bacteriana producida a partir de Corynebacterium diphtheriae. Contiene tres dominios que tienen acciones intracelulares. Los dominios intervienen en la intoxicación de la célula, la unión a la superficie celular y la internalización en endosomas, translocándose al citosol a través de la membrana del endosoma e inhibiendo la síntesis de proteínas celulares. La toxina diftérica es útil en la construcción de herramientas y tratamientos biotecnológicos. Inhibe la síntesis de proteínas al catalizar la adenosina difosfato (ADP)-ribosilación de la proteína diana en eucariotas.

General description

La toxina de la difteria puede utilizarse como el elemento toxina en la construcción de inmunotoxinas para citotoxicidad celular específica. Se ha utilizado el dominio de translocación de las toxinas bacterianas con un mecanismo de escape natural del endosoma en el desarrollo de vectores no víricos eficaces para aplicación en genoterapia.

Physical form

Polvo liofilizado que contiene Tris y EDTA.

Preparation Note

Cada vial, una vez reconstituido a 0,5 ml con agua destilada estéril, contiene ~1 mg de toxina de la difteria en Tris 0,01 M y Na2EDTA 0,001 M, pH 7,5.

    For assays employing low concentration of the toxin, the use of a carrier protein like serum albumin is recommended.

    In the presence of a reducing agent, on SDS gels, this preparation migrates as a major band of approx. mw 63,000 Daltons (corresponding to thte intact toxin) and two faster more lightly stained bands of approx. MW 39,000 and 24,000 Daltons (corresponding to the B and A fragments, respectively). Based on this method, the extent of the degree of nicking in this preparation is estimated to be less than 5%.
