Se utiliza para establecer gradientes de densidad no tóxicos con baja viscosidad y poca osmolaridad que pueden utilizarse para aislar células, orgánulos y virus. Se ha utilizado para separar vesículas de la membrana interna y externa de E. coli. El Percoll es isosmótico en todo el gradiente, no tóxico y fácil de eliminar de los materiales purificados.
Se ha utilizado Percoll® para la preparación del esperma en estudios de los requisitos de energía de los embriones bovinos.

General description

Percoll® es un medio clásico para centrifugación en gradiente de densidad de células, virus y partículas celulares. Percoll consiste en partículas de sílice coloidal de 15– 30 nm de diámetro (23% p/p en agua) que han sido recubiertas con polivinilpirrolidona (PVP). La PVP hace que el producto sea completamente atóxico e ideal para utilizar con materiales biológicos.

    The product information sheet (under Documents, above) has an equation for calculating density of a given solution.

  2. When using Product P1644, Percoll®, what density should I use for my particular system?

    The product information sheet (under Documents, above) has a table of densities for different organelles and cell types.

  3. Is there a "best" salt solution to use with Product P1644, Percoll®?

    Percoll can be used with various salt formulations (PBS, 1.5 M NaCl, HBSS, etc.). Physiological saline and balanced salt solutions can be suggested.  Sucrose is included to prevent aggregation.  You can find detailed preparation instructions on the product information sheet (under Documents, above) for this product.

  4. Can Percoll®, Product P1644, be autoclaved?

    Percoll can be autoclaved, but this must be done without any salts or sucrose present.
