General description
Natural killer cell receptor 2B4 (UniProt: Q9BZW8; also known as CD244, NK cell activation-inducing ligand (NAIL), NK cell type I receptor protein 2B4 (NKR2B4; h2B4), SLAM family member 4 (SLAMF4), Signaling lymphocytic activation molecule 4) is encoded by the CD244 (also known as 2B4) gene (Gene ID: 51744) in human. CD244 (2B4) is a SLAM family receptor broadly expressed on NK cells, subsets of CD8+ αβ and γδ T cells, dendritic cells, and myeloid-derived suppressor cells (MDSCs), where it binds CD48 and signals via four ITSM motifs that recruit SH2 adaptors to drive activating (SAP) or inhibitory (SHP1/SHP2/SHIP-1; context-dependent EAT-2) outcomes; the net signal is governed by receptor density and adaptor availability, with high CD244 and/or limited SAP favoring inhibition. In the tumor microenvironment, CD244 signaling is predominantly inhibitory—promoting NK and CD8+ T-cell exhaustion and dampening cytokine production, while in DCs and Gr-MDSCs it aligns with increased immunosuppressive function—mechanisms often dependent on CD244–CD48 interactions with tumor-associated immune cells. These insights support therapeutic CD244 blockade to reinvigorate exhausted effectors, enhance proinflammatory DC activity, and diminish MDSC-mediated suppression; supportive preclinical data, the grossly normal phenotype of CD244−/− mice, and evidence that leukemia cells can depend on CD244 signaling, collectively underscore CD244 as a promising immuno-oncology target.

Preparation Note
1.0 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
