General description

Neural cell adhesion molecule 1 (UniProt: P13596 ; also known as N-CAM-1, NCAM-1, CD56) is encoded by the Ncam1 (also known as Ncam) gene (Gene ID: 24586) in rat. Neural cell adhesion molecule 1 (NCAM1) is a glycoprotein belonging to the immunoglobulin superfamily. Structurally, NCAM1 is composed of three major isoforms NCAM-120, NCAM-140, and NCAM-180 distinguished by their molecular weight and subcellular localization. These isoforms contain extracellular immunoglobulin-like domains that mediate homophilic and heterophilic interactions, a transmembrane domain, and a cytoplasmic tail that participates in intracellular signaling. NCAM1 functions as both a cell adhesion molecule and a signal transducing receptor, playing critical roles in cell-cell adhesion, neuronal migration, synaptic formation, and cytoskeletal organization. NCAM1 is highly expressed in the central nervous system (CNS), peripheral nerves, adrenal glands, and immune cells, including natural killer (NK) cells. Its expression is also detected in pulmonary arterial smooth muscle cells, where it modulates cellular proliferation and migration under pathological conditions such as pulmonary hypertension. Furthermore, NCAM1-induced signaling pathways are implicated in neuronal survival, astrocyte proliferation, and neurite outgrowth, highlighting its impact on neurodevelopment and repair processes. This protein interacts with a variety of partners, including extracellular matrix proteins, growth factors, and other cell adhesion molecules, via its extracellular modules. These interactions trigger downstream signaling cascades, such as the activation of mitogen-activated protein kinases (MAPKs) and alterations in cytoskeletal dynamics. Additionally, NCAM1 has been identified as an immune checkpoint molecule in mollusks, where it exhibits immunoreceptor tyrosine-based inhibition motifs (ITIMs) in its intracellular domain. This suggests a unique immunosuppressive role in innate immunity, extending its functional versatility beyond vertebrates. Aberrant expression and mutations in NCAM1 are associated with various diseases, including neurodegenerative disorders, cancer, and pulmonary hypertension. Its role in promoting tumor growth and metastasis, as well as its involvement in immune system modulation, makes NCAM1 a potential therapeutic target for clinical interventions.

Preparation Note

0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
