General description
160 kDa
AS160 (Akt Substrate of 160 kDa, TBC1D4) is a Rab-GAP protein that regulates the trafficking of GLUT4 (glucose transporter 4) to the plasma membrane. AS160 is known to be phosphorylated on at least 8 residues following stimulation from IGF-1, EGF, PMA, and AICAR (Chen, S., 2008). Thr642 is phosphorylated by Akt in response to IGF-1, while Ser341 and Thr642 of AS160 are phosphorylated by RSK (p90 ribosomal S6 kinase) in response to PMA. Ser588 has been shown to be phosphorylated by AMPK in vitro, though other kinases might be responsible as well. The phosphorylation site, Ser666, does not contain the Akt consensus sequence. Its phosphorylation is suppressed by AICAR and by combined mutations of TOS (mTOR signaling)-like sequence (FEMDI) and rapamycin (Geraghty, K.M., 2007). Additionally, its phosphorylation is markedly increased in response to IGF-1 and this phosphorylation is blocked in response to the PI3 Kinase inhibitor LY294002 (Geraghty, K.M., 2007).
