General description

A MAC (Mitochondrial Apoptosis-induced Channel) inhibitor iMAC1 (Cat. No. 196805 ) structural analog that is reported to be less potent than iMAC1 in blocking Bid-induced cytochrome c release in cell-free mitochondria preparations (40% vs. 77% inhibition with 5 µM respective inhibitor), but more effective than iMAC1 in inhibiting STS- (Cat. No. 569396 & 569397) induced cytochrome c release in intact FL5.12 cells (Complete vs ~35% inhibition with 5 µM respective inhibitor), presumably due to an enhanced permeability. iMAC2 is also shown to be a superior blocker of MAC-mediated current flow in reconstituted mitochondrial outer membrane patches from apoptotic FL5.12 cells (IC50 = 28 vs 300 nM in the case of iMAC2 and iMAC1, respectively) and protect FL5.12 from STS-induced apoptosis (IC50 = 2.5 µM).
