General description
A cell-permeable di-coumarin compound that selectively inhibits γ-secretase-mediated production of Aβ42 over Aβ38, Aβ40, and Notch1 both in cell-free APP cleavage assays (IC50 = 0.07, 0.71, 0.31, and 1.77 µM, respectively) and in human and murine cultures (100% blockage of Aβ42 production and no effect against the rest at 30 µM) by altering the S1′ and S3′ pockets within the presenilin substrate binding site via an allosteric, non-substrate-competitive mechanism.
