General description

A peptide that contains the N-terminal seven residues of Smac (Second Mitochondria-derived Activator of Caspases, also known as DIABLO) and promotes procaspase-3 activation at around 10 µM. Reported to bind to the BIR2 and BIR3 (baculovirus IAP repeat) domains of XIAP (X-linked inhibitor of apoptosis proteins) with a Kd ~800 nM. Promotes the catalytic activity of caspase-9 in the presence of XIAP, suggesting that Smac competes with caspase-9 for binding to the same pocket on the surface of XIAP.

Biochem/physiol Actions

Cell permeable: no
Kd ~800 nM in binding to the BIR2 and BIR3 (baculovirus IAP repeat) domains of XIAP (X-linked inhibitor of apoptosis proteins)
Primary Target
Promotes the catalytic activity of caspase-9 in the presence of XIAP
Product does not compete with ATP.
Reversible: no

Packaging

Packaged under inert gas

Physical form

Supplied as a trifluoroacetate salt.
