Protein tyrosine phosphorylation is a key element in several membrane signal transduction systems. There are two general classes of tyrosine kinases: the receptor tyrosine kinases and the receptor-associated tyrosine kinases. The receptor tyrosine kinases possess an extracellular ligand binding domain and an intracellular catalytic domain with intrinsic tyrosine kinase activity. The receptor-associated tyrosine kinases transmit signals from the membrane by interacting with the cytoplasmic domain of membrane proteins.



The design of specific inhibitors of tyrosine kinases is important for both fundamental research and for developing therapeutic strategies for the treatment of cancer, atherosclerosis, psoriasis, septic shock, and other disorders where increased tyrosine kinase activity has been reported.
