General description

Vesicle-associated membrane protein 7 (VAMP-7), also known as Synaptobrevin-like protein 1 or Tetanus-insensitive VAMP (Ti-VAMP), and encoded by the gene name VAMP7 or SYBL1, is a member of the soluble N-ethylmaleimide-sensitive factor attachment protein receptor (SNARE) family. VAMP-7 is localized to late endosomes and lysosomes and is thought to mediate the fusion of endosomes to their target lysosomes as well as other exocytosis events during phagocytosis and neuritogenesis. VAMP-7 interacts with the VPS9 ankyrin repeat protein VARP, a protein that localizes to early endosomes and thought to regulate endosome dynamics. Together with CD82, VAMP-7 can modulate the signaling of EGFR by regulating its endocytosis from the plasma membrane.

KLH-conjugated linear peptide corresponding to the cytoplasmic domain of human VAMP-7.

Application

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected VAMP-7 in HeLa and PC12 cells.
Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected VAMP-7 in human kidney, human thalamus, and human cerebral cortex tissue.

This Anti-VAMP-7 Antibody is validated for use in Western Blotting and Immunocytochemistry and Immunohistochemistry for the detection of VAMP-7.

Biochem/physiol Actions

This antibody detects VAMP-7a (isoform 1) and is expected to cross-react with VAMP-7b (isoform 2), but not VAMP-7c (isoform 3).

Purified rabbit polyclonal in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.

Preparation Note

Stable for 1 year at 2-8°C from date of receipt.

Analysis Note

Evaluated by Western Blotting in rat brain tissue lysate.

Western Blotting Analysis: 2.0 µg/mL of this antibody detected VAMP-7 in 10 µg of rat brain tissue lysate.
