General description
Daple (UniProt Q9P219; also known as Hook-related protein 2, HkRP2, hDaple, Coiled-coil domain-containing protein 88C, Dvl-associating protein with a high frequency of leucine residues) is encoded by DAPLE (also known as KIAA1509, SCA40) gene (Gene ID 440193) in human. It acts as a negative regulator of the canonical Wnt signaling pathway and acts downstream of DVL to inhibit CTNNB1/beta-catenin stabilization. Daple has a PDZ-domain binding motif in its C-terminus with which it interacts with the DVL protein. Daple also contains a Gα-binding and activating motif, which activates Gαi proteins and an adjacent domain that directly binds frizzled receptors, thereby linking Wnt stimulation to G protein activation. This triggers non-canonical Wnt responses and may suppress the beta-catenin/TCF/LEF pathway and tumorigenesis, but can enhance PI3K-Akt and Rac1 signals by releasing beta/gamma subunits and may contribute to tumor cell invasiveness. Mutations in this DAPLE gene are known to cause autosomal recessive, primary non-syndromic congenital hydrocephalus.

This rabbit polyclonal antibody targets the region between the coiled-coils and DVL1-/PDZ-binding domain. Target region is present in all three human Daple spliced isoforms reported by UniProt (Q9P219). Specificiy of target band detection was verified using lysates from Daple shRNA-treated cells.
