General description

Daple (UniProt Q9P219; also known as Hook-related protein 2, HkRP2, hDaple, Coiled-coil domain-containing protein 88C, Dvl-associating protein with a high frequency of leucine residues) is encoded by DAPLE (also known as KIAA1509, SCA40) gene (Gene ID 440193) in human. It acts as a negative regulator of the canonical Wnt signaling pathway and acts downstream of DVL to inhibit CTNNB1/beta-catenin stabilization. Daple has a PDZ-domain binding motif in its C-terminus with which it interacts with the DVL protein. Daple also contains a Gα-binding and activating motif, which activates Gαi proteins and an adjacent domain that directly binds frizzled receptors, thereby linking Wnt stimulation to G protein activation. This triggers non-canonical Wnt responses and may suppress the beta-catenin/TCF/LEF pathway and tumorigenesis, but can enhance PI3K-Akt and Rac1 signals by releasing beta/gamma subunits and may contribute to tumor cell invasiveness. Mutations in this DAPLE gene are known to cause autosomal recessive, primary non-syndromic congenital hydrocephalus.
~260 kDa observed. 228.2/57.53/49.25 kDa (isoform 1/2/3) calculated. Uncharacterized bands may be observed in some lysate(s).

Immunogen

His-tagged recombinant human Daple fragment from the region between the coiled-coils and DVL1-/PDZ-binding domain.

Application

Detect Daple using this rabbit polyclonal Anti-Daple, CT, Cat. No. ABS515, validated for use in Immunocytochemistry and Western Blotting.
Immunocytochemistry Analysis: A 1:300 dilution from a representative lot detected exogenously expressed human Daple in paraformaldehyed-fixed HEK293 transfectants (Courtesy of Pradipta Ghosh, M.D, UC San Diego, U.S.A.).

Western Blotting Analysis: A 1:500 dilution from a representative lot detected a ~260 kDa target band in lysate from control shRNA-, but not Daple shRNA-, treated HeLa cells (Courtesy of Pradipta Ghosh, M.D, UC San Diego, U.S.A.).

Western Blotting Analysis: A representative lot detected exogenously expressed human Daple in lysates from various transfected cells (Aznar, N., et al. (2015). Elife. 4:e07091).

Biochem/physiol Actions

This rabbit polyclonal antibody targets the region between the coiled-coils and DVL1-/PDZ-binding domain. Target region is present in all three human Daple spliced isoforms reported by UniProt (Q9P219). Specificiy of target band detection was verified using lysates from Daple shRNA-treated cells.

Physical form

Affinity purified.
Purified rabbit polyclonal antibody in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.

Preparation Note

Stable for 1 year at 2-8°C from date of receipt.

Analysis Note

Evaluated by Western Blotting in lysate from Daple-transfected COS7 cells.

Western Blotting Analysis: A 1:50,000 dilution of this antibody detected the expression of Myc-tagged full-length human Daple in lysate from transfected COS-7 cells.

Other Notes

Concentration: Please refer to lot specific datasheet.
