General description
Ash1L, also known as Histone-lysine N-methyltransferase ASH1L, or ASH1-like protein (huASH1), Absent small and homeotic disks protein 1 homolog, or Lysine N-methyltransferase 2H, and encoded by the gene ASH1L/KIAA1420/KMT2H, is a histone methyltransferase that specifically methylates Lys-36 of histone H3 (H3K36me). ASH1L is one of several H3K36-specific methyltransferases. Others include HYPB, NSD1 and NSD2 proteins. ASH1L′s activity is regulated by many processes but one interesting one is the inhibition of ASH1L by the presence of ubiquitinated histone H2a (ubH2a). All of the H3K36-specific methyltransferases, including ASH1L, HYPB, NSD1 and NSD2 appear to be inhibited by ubH2A, whereas the other histone methyltransferases, including PRC2, G9a and Pr-Set7 are not affected by ubH2A. This finding helps to partially explain how and why particular histone H3 proteins get methylated while others do not and thus provide some insight into is the mechanisms involved in polycomb chromatin structure and its effects on gene expression or silencing. Ash1L is localized to the nucleus and tight junctions and is widely expressed.
