Enzyme Immunoassay: A representative lot of this antibody detected rat and mouse collagen Type I, but not human collagen in ELISA application.

Dot Blot Analysis: A 1:500 dilution from a representative lot detected rat and mouse tail collagen, but not human placenta collagen.

Peptide Inhibition Assay: Target band detection in rat skin tissue lysate was prevented by pre-blocking of a representative lot of this antibody with rat tail collagen type I.

Western Blotting Analysis: A 1:2,000 dilution from a representative lot detected Collagen Type I in rat skin tissue lysate.

General description

Collagen alpha-1(I) chain (UniProt: P02454; also known as Alpha-1 type I collagen) is encoded by the Col1a1 gene (Gene ID: 29393) in rat. Collagen alpha-1(I) chain (Col1a1) is a fibril-forming structural protein predominantly found in connective tissues, including bone, cornea, dermis, and tendon. Type I collagen is composed of two alpha-1 chains and one alpha-2 chain, forming a triple helical structure that provides tensile strength and mechanical stability to tissues. It functions as an extracellular matrix constituent, conferring rigidity and facilitating cellular responses to mechanical stimuli and signaling pathways such as collagen-activated tyrosine kinase receptor signaling. It is localized in the extracellular space and secretory granules, playing a key role in extracellular matrix assembly and maintenance. The expression of Col1a1 is regulated under various physiological and pathological conditions. It is upregulated during tissue remodeling processes, such as wound healing and fibrosis, and plays a role in pathological states, including myocardial infarction where it contributes to infarct healing through extracellular matrix remodeling. Aberrant expression or mutations in the Col1a1 gene are implicated in diseases such as osteogenesis imperfecta, Ehlers-Danlos syndrome, and idiopathic osteoporosis. Increased Col1a1 expression has been linked to processes like epithelial-to-mesenchymal transition (EMT), cancer progression, and invasion in conditions such as cholangiocarcinoma, making it a potential biomarker for disease severity and prognosis. Type I collagen is widely used in cartilage tissue engineering. (Ref.: Hou, L. et al., (2021) J Cancer. 12(19), 5723-5731; Jiang YH, et al., (2022) Am J Transl Res., 14(2), 1146-1159).
