General description
TNF receptor-associated factor 1 (UniProt: Q13077; also known as Epstein-Barr virus-induced protein 6, TRAF1) is encoded by the TRAF1 (also known as EB16) gene (Gene ID: 7185) in human. TRAF1 is homodimeric pro-survival adapter molecule that regulates the activation of NF-kB and JNK. It can also exist as a heterotrimer with TRAF2. It plays a role in the regulation of cell survival and apoptosis. The heterotrimer form is part of a E3 ubiquitin-protein ligase complex that promotes ubiquitination of target proteins, such as MAP3K14. The TRAF1/TRAF2 complex recruits the antiapoptotic E3 protein-ubiquitin ligases BIRC2 and BIRC3 to TNFRSF1B/TNFR2. The C-terminal region of TRAF1 contains the TRAF domain (aa 266-412) that binds to receptor cytoplasmic domains. It also has a coiled coil domain (aa 182-264) that mediates its homo- and heterotrimerization. TRAF1 is also reported to essential for memory T cell survival. TRAF1 protein level is significantly reduced in HIV-specific CD8 T cells with progression of infection. Its levels can increase following treatment with IL-7. Hence, TRAF1 levels could serve as a useful parameter in monitoring effectiveness of anti-HIV therapies. (Ref.: Wang, C., et al. (2018). J. Immunol. 200(2); 558-564).
