General description
Genome polyprotein (UniProt: Q32ZE1) from Zika virus strain Mr 766 is a large molecular weight protein. In Zika virus surface proteins are arranged in an icosahedral-like symmetry. Attachment of the viral envelope protein E to host receptors mediates internalization into the host cell by apoptotic mimicry. Following the fusion of virus membrane with host endosomal membrane, RNA genome is released into the cytoplasm. Polyprotein components include Protein C homodimerizes and plays a role in virus budding by binding to membrane and gathering the viral RNA into a nucleocapsid that forms the core of a mature virus particle. The Pr peptide prevents premature fusion activity of the envelope proteins in trans golgi by binding to envelope protein E (at pH 6.0). The PrM peptide serves as a chaperone for envelope protein E during intracellular virion assembly. Polyprotein also has a RNA-directed RNA polymerase NS5 that replicates the viral (+) and (-) genome and methylates viral RNA cap at guanine N-7 and ribose 2 -O positions. The non-structural protein 1 can form homodimers and homohexamers is involved in immune evasion, pathogenesis and viral replication and non-structural protein 2A functions in virion assembly and antagonizes the host immune response. The non-structural protein 2B is essential for the serine protease function of NS3. The serine protease NS3 has three different enzymatic activities - serine protease, NTPase and RNA helicase. The non-structural protein 4A of virus regulates the ATPase activity of the NS3 helicase while the non-structural protein 4B inhibits interferon-induced host STAT1 phosphorylation and its nuclear translocation. The 2k peptide serves as a signal peptide for NS4B and is required for the interferon antagonism. Clone ZV-54 is reported to neutralize infection of African, Asian, and American strains of Zika virus to varying degrees. (Ref.: Zhao, H et al. (2016). Cell 166(4); 1016-27).
