General description

Calmodulin-like protein 5 (UniProt Q9NZT1; also known as Calmodulin-like skin protein) is encoded by the CALML5 (also known as CLSP) gene (Gene ID 51806) in human. Epidermal differentiation and barrier function depend on many calcium-regulated proteins and enzymes. CALML5/CLSP is expressed in the stratum granulosum and lower layers of the stratum corneum of the skin, where it modulates the function of calcium-dependent proteins, such as transglutaminase 3. CALML5/CLSP level is upregulated in granular and spinal keratinocytes during the later stages of wound healing in human epidermis and eventually returns to basal level once the wound is closed and barrier function has been restored. In the hyperproliferative skin disorders psoriasis and atopic dermatitis (AD), the permeability barrier is disturbed, causing an increased CLSP built-up as a result of decresed CLSP degradation due to an altered calcium gradient.
