General description

α-Synuclein is a small, soluble protein predominantly found in the presynaptic terminals of neurons. It actively participates in synaptic function by regulating neurotransmitter release and maintaining synaptic integrity. Additionally, α-synuclein influences the dynamics of synaptic vesicles and interacts with various proteins involved in synaptic transmission. In neurological disorders, particularly neurodegenerative diseases like Parkinson′s disease, α-synuclein misfolds and aggregates into insoluble fibrils, forming Lewy bodies. These aggregates disrupt cellular function, leading to neuronal toxicity and death. The accumulation of α-synuclein also triggers neuroinflammation by activating microglial cells, which can further contribute to neurodegeneration.

Analysis Note

  • Sensitivity: LLOQ 1,950 fg/mL* (SMCxPRO® Instrument)
  • Standard Curve Range: 490-500,000 fg/mL
  • Intra-assay CV: <15%
  • Inter-assay CV: <20%
  • Linearity: Serum: 113% Plasma: 118% CSF: 102%
  • Recovery: Serum: 96% Plasma: 96% CSF: 96%
*Indicative LLOQ created from average of 14 independent in house runs, where >80% met same value defined with CV & Recovery +/- 20%. Refer to CoA for lot specific performance.

Other Notes

Although Total α-Synuclein was detectable in non-human primate (rhesus macaque and cynomolgus monkey), feline, rat, mouse, and canine samples, the assay has not been verified for these species.
