General description
Des-Ile(12)-Tirzepatide is a deletion impurity of Tirzepatide—a dual GIP/GLP-1 receptor agonist—in which Isoleucine at position 12 (API numbering; position 12 in GIP/GLP-1 numbering) is absent, yielding a 38-residue peptide. Tirzepatide contains the non-standard residues Tyr1 and Aib2 at the N-terminus, and carries an AEEAc-AEEAc-γGlu-C20 fatty diacid conjugate at Lys19 (API numbering). The deletion of Ile12, located within the central helical segment critical for receptor binding, may significantly disrupt the conformational alignment required for dual GIP and GLP-1 receptor engagement. This impurity may arise during SPPS as a deletion sequence or from incomplete coupling of the sterically hindered Ile residue.
