General description
TAR DNA-binding protein 43 (UniProt: Q13148; also known as TDP-43) is encoded by the TARDBP (also known as TDP43) gene (Gene ID: 23435) in human. TDP-43 is a RNA binding protein that is suggested to be involved in various steps of RNA biogenesis and processing and, is mainly present in nucleus. Structurally, it consists of a N-terminal domain (NTD) and two tandem RNA recognition domains (RRM1 and RRM2), followed by a c-terminal glycine-rich region. RRM1 and RRM2 domains are responsible for binding the GU-repeats on RNA molecule. TDP-43 also contains a nuclear localization sequence and a nuclear export sequence, which permits it to transport mRNAs to the cytoplasm and even to synapses as part of neuronal granules. It is hypothesized that TDP-43 forms oligomeric assemblies that associate with tau. Tau oligomers are reported to modulate TDP-43 aggregation state and cellular localization, and TDP-43 aggregates are detected in Alzheimer s disease (AD) brains. Aggregates of post-translationally modified TDP-43 in cytoplasmic inclusion bodies are also characteristic features in frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS). Studies using various biochemical and biophysical assays that examined cross-seeding property of AD, ALS, and FTD brain-derived TDP-43 oligomers (BDT43Os) on tau aggregation suggest that TDP-43/tau interactions may play a role in these neurodegenerative diseases. Preparation Note

Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
