Friend leukemia integration 1 transcription factor (UniProt: Q01543; also known as Proto-oncogene Fli-1, Transcription factor ERGB) is encoded by the FLI1 gene (Gene ID: 2313) in human. FLI1 transcription factor is a member of the ETS family of transcription factors and plays critical roles in diverse biological processes, including cell proliferation, differentiation, apoptosis, and angiogenesis. FLI1 contains an ETS DNA-binding domain, which enables sequence-specific binding to target genes, and a sterile alpha motif (SAM) domain associated with protein-protein interactions. The protein regulates transcription mediated by RNA polymerase II. It is essential for normal hematopoiesis and vascular morphogenesis but is also implicated in oncogenesis, autoimmune diseases, and inflammatory conditions. RNA-binding protein EWS (EWSR1) is a member of the TET family, a highly conserved group of multifunctional, RNA-binding proteins that play critical roles in transcription regulation, RNA processing, and cellular signaling. Chromosomal translocations (most commonly the translocation t(11;22)(q24;q12), involving the Ewing sarcoma breakpoint region 1 gene (EWSR1) and FLI1 (EWSR1-FLI1 fusion gene) are a hallmark of Ewing sarcoma, driving tumorigenesis by aberrant regulation of genes associated with cell growth and survival. In the EWS-FLI1 fusion protein the RNA binding domains from the C terminus of EWS are replaced by the ETS DNA-binding domain of FLI1. Inhibitors of the EWSR1-FLI1 fusion protein in Ewing sarcoma are under investigation for their ability to mitigate tumor growth. (Ref.: Gong, H., et al., (2023). Cancers. 15(16), 4035; Erkizan H.
