General description
Collagen is classified into a number of structurally and genetically distinct types. We use the nomenclature proposed by Bornstein and Traub. Do not confuse Sigma type designations with recognized collagen classification types.
Collagen is classified into several structurally and genetically distinct types. Type IV collagen is a predominant structural protein of the basement membrane. It is notably characteristic of the Engelbreth-Holm-Swarm (EHS) mouse sarcoma. It is commercially available as matrigel and cultrex. Matrigel is made of type 4 collagen, laminin, heparan sulfate proteoglycan and entactin obtained from engelbreth-holm-swarm murine sarcoma. The target cells for attachment include: epithelial cells, endothelial cells, muscle cells, and nerve cells.
Application
Recommended for use as a cell culture substratum at 6-10 μg/cm2.
Collagen from Engelbreth-Holm-Swarm murine sarcoma basement membrane has been used:
- in adhesion assays.
- in hydrogel fabrication.
- to coat cell culture plates for cell differentiation.
- to treat round plastic coverslips for scanning electron microscopy (SEM) analysis in an interaction study between A. culbertsoni trophozoites and Schwann cells.
Biochem/physiol Actions
Collagen isolated from Engelbreth-Holm-Swarm murine sarcoma basement membrane helps in the development of 3D nodules. It is also used in 2D coatings.
