General description

FGF-basic is one of 23 known members of the FGF family. Proteins of this family play a central role during prenatal development and postnatal growth and regeneration of a variety of tissues, by promoting cellular proliferation and differentiation. FGF-basic is a non-glycosylated heparin binding growth factor that is expressed in the brain, pituitary, kidney, retina, bone, testis, adrenal gland liver, monocytes, epithelial cells and endothelial cells. FGF-basic signals through FGFR 1b, 1c, 2c, 3c and 4. Recombinant human FGF-basic is a 17.2 kDa protein consisting of 154 amino acid residues.

Application

FGF-basic has been used to supplement serum-free DMEM during culture of HeLa cells.

Biochem/physiol Actions

FGF-basic (fibroblast growth factor), also referred to as FGF2, activates the ERK (extracellular signal-regulated kinase) cascade and facilitates self-renewal of hESCs (human embryonic stem cells). FGF2 positively regulates cell survival and/or proliferation. It is found to negatively regulate BMP (bone morphogenetic protein) signaling, a pathway that induces non-neuronal differentiation.

Physical form

Lyophilized from 5 mM Tris, pH 7.6 + 150mM NaCl.

Preparation Note

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8° C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 5% Trehalose) and store in working aliquots at -20° C to -80° C.
