El Dl-ditiotreitol es un componente fundamental utilizado en la preparación de disolución tampón que, a su vez, se utiliza en la preparación citosólica plaquetaria. También es importante en el aislamiento del ARN utilizando sales de guanidinio y homogeneización de tejido y células.
Un reactivo excelente para el mantenimiento de los grupos SH en estado reducido; reduce cuantitativamente los disulfuros. El DTT reduce de manera eficaz los enlaces disulfuro de las proteínas en tampones de muestra antes de la SDS-PAGE. El DTT puede utilizarse también para reducir el puente disulfuro del reticulante N,N′-bis(acriloil)cistamina con el fin de separar la matriz de un gel de poliacrilamida. El DTT es menos mordaz y menos tóxico que el 2-mercaptoetanol. Normalmente se requiere una concentración de DTT siete veces inferior (100 mM) a la concentración utilizada del 2-mercaptoetanol (5 % v/v, 700 mM).

Biochem/physiol Actions

El Dl-ditiotreitol desnaturaliza las proteínas al reducir los enlaces disulfuro a grupos SH. También impide la formación de puentes disulfuro intramoleculares e intermoleculares entre los residuos de cisteína de las proteínas.
