General description
Jagged-1 is a Notch ligand. The Notch signaling system regulates proliferation, differentiation, and apoptosis in many tissues, including hematopoietic tissue. Notch is a family of transmembrane receptors activated by ligands (Jagged-1, Jagged-2, and Delta-1) that are expressed on adjacent cells. Hematopoietic stem cells and early progenitors express Notch-1 and Notch-2, whereas primary stromal cultures have been shown to express Jagged-1, indicating that Jagged-1 may play a role in nurturing progenitor cells within the hematopoietic microenvironment. Cells engineered to express high levels of Jagged-1 caused a four-fold increase in formation of hematopoietic progenitor cell colonies. However, without the presence of other cytokines that stimulate proliferation and maturation, such as IL-3, IL-6, SCF and G-CSF, Jagged-1 actually decreased myeloid colony formation of CD34+ cells, suggesting that Jagged-1 modulates cytokine signals and helps preserve CD34+ cells in an immature state.
Notch signaling is a well conserved among the metazoans and is a regulator of growth, differentiation, and patterning processes. One of the ligands of Notch family receptors is Jagged. Mammalian Jagged family comprises of Jagged-1 and Jagged-2. Jagged proteins are expressed during the critical stages of embryonic and postnatal development. The Notch-Jagged signaling regulates the proliferation of endothelial cells, apoptosis, differentiation of hematopoietic progenitors, skeletal development and formation and differentiation of blood vessels. Notch-jagged pathway components play important role in the pathology of diseases like Alagille syndrome, CADASIL (cerebral autosomal dominant arteriopathy with subcortical infarcts and leukoencephalopathy) and osteosarcoma. Anti-Jagged 1 antibody specifically recognizes rat Jagged-1.

recombinant rat Jagged 1 extracellular domain, expressed in NSO cells.
Application
Anti-Jagged 1 antibody may be used for ELISA at a working antibody concentration of 0.5-1.0 μg/ml. For immunoblotting, antibody concentration of 0.1-0.2 μg/ml is recommended. For flow cytometry, a concentration of 20 μg/ml (10 μL/105 cells to 106 cells) may be used. The antibody is suitable for neutralization assays (ND50 is ~2-10 μg/ml).
