General description
Transient receptor potential cation channel subfamily M member 7 (UniProt: Q923J1; also known as EC:2.7.11.1, Channel-kinase 1, Long transient receptor potential channel 7, LTrpC-7, LTrpC7, Transient receptor potential-phospholipase C-interacting kinase, TRP-PLIK) is encoded by the Trpm7 (also known as Chak, Ltrpc7) gene (Gene ID: 58800) in murine species. It is a bifunctional protein that exhibits both ion channel and kinase activities, essential for various physiological functions in mice. Subcellular localization studies reveal that TRPM7 is a multi-pass membrane protein and is predominantly found at the plasma membrane and within specialized cellular compartments, where it interacts with cytoskeletal proteins, influencing cell morphology and motility. It forms a non-selective cation channel with notable permeability to divalent cations such as Mg²⁺ and Zn²⁺, facilitating their influx into cells, which is critical for processes like embryonic development and cellular signaling. It is ubiquitously expressed across multiple tissues, including brain, skeletal muscle and, with stronger signals in kidney, heart, liver and spleen. Its also expressed in immune cells, indicating its fundamental role in cellular homeostasis and signaling pathways. Dysregulation of TRPM7 has been implicated in various diseases, including neurodegenerative disorders, cancer, and cardiac dysfunction, highlighting its potential as a therapeutic target. Research studies have elucidated the mechanisms by which TRPM7 contributes to neuronal survival and development, as well as its involvement in muscle regeneration and pyroptosis, a form of programmed cell death linked to inflammatory responses. A recent study reported that TRPM7 is crucial for early embryos because it maintains magnesium (Mg2+) levels, protects against oxidative stress, and promotes the proper gene activity required for development and cell differentiation before the blastocyst stage.
